You can also take a bus from ISBT Kashmere Gate or Mussoorie to Harsil and then hire a private taxi from there to Hanuman Chatti. If you're already in Uttarakhand, you can take a train or bus till Haridwar and then another bus or shared jeep till Rishikesh. From Rishikesh, there are regular buses and shared jeeps going till Uttarkashi. From Uttarkashi, you can again take a bus or shared jeep till Hanuman Chatti which is the starting point for the trek.The trek from Hanuman Chatti to Yamunotri is around 13 km. Devotees can be complete this in 4-5 hours. The first 3 kilometers of the trek are easy but after that, it becomes quite steep.
